Self-Administration of JWH-018 A Synthetic Cannabinoid in Experimentally Naïve Rats

چکیده

A recent study by Dr. Maria Antonietta De Luca demonstrated intravenous (IV) self-administration responding (nose-poking) for the synthetic cannabinoid JWH-018 [1-pentyl-3-(1-naphthoyl)indole] (Figure 1) in an experimentally naïve, adult rat species [1]. This finding is unexpected since the phytocannabinoid (−)-trans-Δ9tetrahydrocannabinol (Δ9-THC, Figure 1), a primary psychoactive constituent in marijuana, has been reported to not maintain IV selfadministration responding above vehicle levels in rats [2,3] and rhesus monkeys [4-6]. IV self-administration of synthetic cannabinoids is not unprecedented since several synthetic cannabinoids have been found to maintain IV self-administration responding in experimentally naïve rats [1,7-10], and mice [11-14]. However, the finding by Dr. De Luca is important because JWH-018 has been frequently found in K2/Spice preparations [15-17] and there continues to be an increase in the abuse and non-medical use of various synthetic cannabinoids worldwide [15-17]. Further, the use of marijuana has been recently legalized in two states of the U.S.

Repeated Exposure to the “Spice” Cannabinoid JWH-018 Induces Tolerance and Enhances Responsiveness to 5-HT1A Receptor Stimulation in Male Rats

Naphthalen-1-yl-(1-pentylindol-3-yl)methanone (JWH-018) is a synthetic compound found in psychoactive "spice" products that activates cannabinoid receptors. Preclinical evidence suggests that exposure to synthetic cannabinoids increases 5-HT2A/2C receptor function in the brain, an effect which might contribute to psychotic symptoms. Here, we hypothesized that repeated exposures to JWH-018 would...

Synthetic cannabinoid JWH-018 and psychosis: an explorative study.

BACKGROUND Aroma, Spice, K2 and Dream are examples of a class of new and increasingly popular recreational drugs. Ostensibly branded "herbal incense", they have been intentionally adulterated with synthetic cannabinoids such as JWH-018 in order to confer on them cannabimimetic psychoactive properties while circumventing drug legislation. JWH-018 is a potent cannabinoid receptor agonist. Little ...

Phase I Hydroxylated Metabolites of the K2 Synthetic Cannabinoid JWH-018 Retain In Vitro and In Vivo Cannabinoid 1 Receptor Affinity and Activity

BACKGROUND K2 products are synthetic cannabinoid-laced, marijuana-like drugs of abuse, use of which is often associated with clinical symptoms atypical of marijuana use, including hypertension, agitation, hallucinations, psychosis, seizures and panic attacks. JWH-018, a prevalent K2 synthetic cannabinoid, is structurally distinct from Δ(9)-THC, the main psychoactive ingredient in marijuana. Sin...

Blood synthetic cannabinoid concentrations in cases of suspected impaired driving.

Twelve cases of suspected impaired driving are discussed in which the drivers who subsequently tested positive for synthetic cannabinoid drugs underwent a psychophysical assessment. The attitude of the drivers was described as cooperative and relaxed, speech was slow and slurred and coordination was poor. Pulse and blood pressure were generally elevated. Horizontal gaze nystagmus was assessed i...

Validation of a novel immunoassay for the detection of synthetic cannabinoids and metabolites in urine specimens.

Synthetic cannabinoid drugs do not cross react on traditional marijuana immunoassay tests, preventing their use in large scale drug screening programs. This paper describes the validation and performance characteristics of two enzyme linked immunosorbent assays designed to detect the use of two common synthetic cannabinoids in urine, JWH-018 and JWH-250. The JWH-018 assay has significant cross-...
